Neocarzinostatin, discovered in 1964, is a naturally occurring antitumor agent. With its unique enediyne chromophore, it was the first protein antibiotic ever used clinically and has become the prototypical anticancer agent in its class. This volume presents a comprehensive treatment of the past, present, and future development of neocarzinostatin, from the earliest findings to the most up-to-date research. Included are descriptions of the general characteristics of neocarzinostatin, the amino acid sequence and chemical structure of the protein portion and the chromophore, two-dimensional NMR study, X-ray crystallography, the mode of action at the molecular level, and the immunopotentiating effect. Clinical effects and side effects are also discussed, as is development of a polymer conjugate for the polymer drug SMANCS and its clinical application.
